I closed and restarted XL4.2.1 and it played once fine. When to the Sequencer and made one change and rendered it. then to the Layout to see what it looked like.. no joy. The play button turns to the pause symbol like it is playing but it isn't..
Any ideas?

Must save, not just render.
The layout Play button is playing the FSEQ file, not what is in the Sequence tab.

Actually the Layout page will now play what you've rendered I just tested it. The confusion usually comes from people trying to use the playback controls up on the toolbar but the Layout page still operate off its own playback controls on the page itself.

The playback stuff on the Layout page is mostly just there for folks that are using the Convert tab to make sure the data got converted more or less correctly. Anyone using the Sequence tab to actually sequence is likely better off just pulling up the Whole House preview pane and using the play controls in the toolbar and not use the stuff on the Layout tab.

I have tried save and render and I am using the play button in the Layout page. It works once, like I said.. then when I click on it, it looks like it is playing, as in it changes to the pause symbol ( ll ), but it isn't. When this happens, I noticed that in the setup tab I can no longer add anything. It greys out all the buttons but the Save. Clicking on that yields nothing. I have to restart XL in order to clear it up.

Sounds like you didn't wait for the render to finish before you tried to play. Kinda hard for us to figure out everything we need to block you from doing.

I am pretty sure I waited long enough, but to be honest I never thought about that since I am just playing with about 20 sec worth of effects (just starting this sequence). Let me see if that is indeed my issue now that I know it could be. Perhaps I am interrupting the rendering process and that is corrupting the file..

Look in the bottom left corner there should be a status message that says something like "Rendered in 34.2 seconds".

I remembered that once you mentioned it..
it renders in less than 3 sec.. I kinda thought so.. but as of right this minute today, no issues... but have not started program changes yet.. I will add more later
