Hi All,
I'm adding on to the display and need to add some E1.31universes. It's been a couple of years since I set up initially so I don't remember what the term "one output" in the E1.31 setup box is referring to. When would you want to 'tick' the radio button to select this?

I'm pretty sure that means when you are outputting multiple universes on one SPI output. It would look something like:
Universes 37-38 510x2

SPI is the way you talk to pixels. (And many other things). Things like WS2811 and TM1804 are pixel chip designs that accept Serial data (one data bit at a time) at high speed, then reassemble into bytes that can be executed. It is why pixels can work with a single data line. Serial Peripheral Interface. The USB on computers is similar.
