Title: Play and stop buttons in the layout tab.

Version 2016.12 the play button will start the sequence playing in the layout tab after that you can't stop it or Paus. After it's played once both buttons become inactive

I have the same problem. Anybody have a reason for why this happens?

I think nobody responded because we've talked about it so many times before.  Those buttons got broken in recent changes but they are being deleted soon anyways so just don't use them.
